GLP-1 / GIP Dual Agonist Once-Weekly Subcutaneous Pharmaceutical Grade

Overview

AFIRA Tirzepatide 40 mg / 3 ml is a pre-filled multi-dose pen containing 40 mg of high-purity tirzepatide in 3 ml sterile liquid (13.33 mg/ml). Tailored specifically for initiation and early titration regimens (2.5 mg and 5.0 mg weekly schedules), it provides multi-week continuous coverage in a single sleek pen device.

Strictly for certified healthcare practitioners and clinical facilities

Dual Incretin Mechanism: GLP-1 + GIP Agonism

Tirzepatide is a first-in-class dual glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptor agonist. By simultaneously activating both incretin pathways, it exerts synergistic metabolic control: promoting glucose-dependent insulin secretion from pancreatic beta cells, suppressing inappropriate glucagon release, slowing gastric motility, and stimulating central hypothalamic satiety circuits to curb appetite and enhance peripheral insulin sensitivity.

Dual Incretin Mechanism of Action (GLP-1 & GIP Synergy)
Dual Incretin Mechanism of Action • Synergistic GLP-1 & GIP receptor activation across brain, pancreas, stomach, and adipose tissue.

Administration

Multi-dose applicator with click dial selector. Affix a new sterile needle prior to injection and administer subcutaneously at 90° into abdominal or thigh adipose tissue.

Clinical Dose Titration & Escalation Protocol

To ensure gastrointestinal tolerability, Tirzepatide administration must follow a stepwise escalation protocol. Treatment initiates at a low starting dose of 2.5 mg subcutaneously once weekly for the initial 4 weeks (adaptation phase). The dose is then increased to 5.0 mg once weekly. Subsequent increments of 2.5 mg may occur at minimum 4-week intervals based on clinical assessment and tolerance, up to the maximum dose of 15.0 mg weekly.

Clinical Titration and Escalation Schedule
Standard Once-Weekly Subcutaneous Escalation Schedule • Minimum 4-week adaptation periods between dose tiers.
